No genes have been identified that explain a major proportion of the heritability of most health outcomes (1). Several vertebrate studies have demonstrated effects of generation 1 nutrition on generation 3 phenotypes (2), and recent work in epigenetics and developmental origins of health and disease has increased interest in transgenerational effects in humans. These effects could be examined in a three-generation cohort; however, best-practice methods for analyzing such multigenerational data, with the associated confounding and selection bias, have not been established.
