Abstract New β-branched glucomannan (TOPL) with an average molecular weight (4.29 × 104 Da) was purified from dandelion leaves for the first time. TOPL possesses a potent IC50 against non-small lung carcinoma (NSCLC); A549 and H460 cell lines (4.01 and 6.57 μg/ml), respectively. To improve the drug-like properties of TOPL, enhance solubility, and counteract its poor bioavailability, the study aimed to design nano-functional particle (nTOPL) by using different nanocarriers such as liposomes and solid lipid nanoparticles (SLNs). Moreover, nTOPL exerted a marked cytotoxic effect against A549 (2.2 μg/ml), H460 (1.7 μg/ml), with minimal toxicity against normal WI-38 cells (18.0 μg/ml). Besides, nTOPL promoted apoptosis and induced arrest in the S-phase, G2/M phase, with up-regulation in Beclin-1, Bax thresholds, and contritely inhibition of Bcl-2 expression. In conclusion, the study implied that nTOPL is a new mannan-lipid nanoparticle that could provide a future promising candidate as a functional food and alternative medicine in lung cancer.
