Three undescribed compounds and nine known compounds were isolated from the Euphorbia fischeriana Steud. Their structures were elucidated by extensive analysis of the 1D and 2D NMR spectra and HRESIMS data. The absolute configurations of two undescribed diterpenoids were determined by comparing their experimental and calculated ECD data. To further understand the antitumor effects of E. fischeriana, we tested the antiproliferative activities of these compounds against AGS, Hep-G2 cell lines in vitro using CCK-8 assays. The results showed that both types of compounds had significant antiproliferative effects, among which a diterpenoid with a galloyl moiety (IC50 = 7.03 μM) was the most significant one that could be regarded as a potential leading compound against liver cancer for further comprehensive study.
