their exhausted T cells revitalized to attack the tumor. Blocking other checkpoints is an obvious approach. Enter the next wave of checkpoints: Lag3, T-cell immunoglobulin and Tim-3, and T-cell immunoglobulin and ITIM domain (TIGIT). They’re all co-expressed with PD-1 on exhausted T cells, and targeting any of them in combination with PD-1 blockade in mouse models is very effective at reinvigorating T cells and eliminating mouse tumors (Eur. J. Immunol. 45, 1892–1905, 2015). These checkpoints all signal differently, have Industry ‘road tests’ new wave of immune checkpoints
