Autoimmune diseases are characterized by the immunemediated attack on seemingly healthy tissue. This attack is caused by a misdirected immune response to self-antigen. A key element of most autoimmune disorders is the recognition by T lymphocytes of specific antigenic peptides loaded onto major histocompatibility complex molecules and known as epitopes. There is firm consensus in the field that inducing immunological tolerance to disease-relevant epitopes would be the ideal approach to treating autoimmunity. In PNAS, Postigo-Fernandez et al. (1) describe their systematic approach to testing this strategy in a mouse model for autoimmune (type 1) diabetes with a DNA vaccination platform they recently developed.
