Significance In this study, we used an efficient and simple synthetic phage engineering platform to generate tailor-made and biologically contained phages for the treatment of drug-resistant and difficult-to-treat bacterial infection. The biocontained phages were genetically virion gene deficient but phenotypically functional and rescued septic mice in a manner similar to that of its parent wild-type phage without producing progeny. Our platform enabled the rebooting of custom-made and biocontained phages from diverse phage families, accelerating a safe synthetic phage therapy for combating drug-resistant superbugs.
