Significance Allelic variation in HHIP confers risk of smoking-related lung disease. We show that HHIP is expressed in the medial habenula, where it regulates the motivational properties of nicotine. These findings have relevance for understanding habenular involvement in nicotine aversion and mechanisms of genetic vulnerability to smoking-related diseases.
