Significance Human complex II deficiency resulting from mutations in the structural subunit genes can lead to metabolic disease or cancer. Here, we have determined the structure of this complex which explains how it effectively transports electrons but also explains why mutations to this complex lead to disease. The study lays a foundation for the development of therapeutic approaches to treat these maladies.
