Significance Using combinatorial loss-of-function CRISPR screens, our study revealed a Bromo- and extra-terminal domain inhibitors (BETi) resistance network in KMT2A-r leukemia. Based on these findings, we have also revealed a corresponding synergy treatment regimen, significantly improving strategies to prevent and treat BETi resistance and relapse that might be seen in the clinic.
