available at http://www.ncbi.nlm.nih.gov/pubmed/30100187 Editorial Comment: The authors aimed to study if specific nucleoporins and nuclear pore complexes-nucleoporin based mechanisms contribute to prostate cancer aggressiveness. They found that the composition of nuclear pore complexes is substantially modified in tumors that progress to advanced disease, and specific nucleoporins enhance the signaling activity of oncogenic and prostate cancer specific transcription factors. In particular the increased expression of nucleoporin POM121 selectively promotes importin mediated nuclear transport of MYC, E2F1, AR and GATA2, which propel tumor initiating, proliferation and survival properties of prostate cancer cells. It is noteworthy that genetic and pharmacological inhibition of the POM121-importin b axis signifcantly diminishes these biological properties. Collectively these results mechanistically define the role of the nuclear pore complexes in the progression of prostate cancer into an aggressive lethal state and identify a potential therapeutic strategy for this disease.
