. Sorghum primary branch length of panicle (PBL) is important for sorghum grain dehydration and yield. In this study, F2 segregating population was constructed using significantly different KY133B and KY123B as parents, and sorghum primary branch length of panicle was preliminarily located by BSA-seq (bulked segregant analysis - sequencing). Through association analysis by ED algorithm and SNP-index (or InDel-index) algorithm, the association region was finally located on chromosome 1, and the total length of association regions was 3.851 Mb, containing 203 genes. By joint transcriptomic analysis, 21 differentially expressed genes were found in the associated regions. Three candidate major genes of primary branch length ( LOC8084886 ; LOC110430248; LOC110431612 ) were inferred by comprehensively analyzing gene functional annotation, Log 2 FC value of differentially expressed genes, TPM of each sample between parents, etc. In this study, based on BSA-seq and transcriptome analysis, sorghum primary branch length major genes were quickly located to lay a foundation for subsequent gene mining.
