A simple and rapid liquid chromatographic method is presented for the determination of morphine HC1, ketamine HC1 and droperidol in 0.9% sodium chloride injection stored in polyvinyl chloride (PVC) infusion bags. Assay was performed on a pre-packed HPLC Hypersil BDS C18 3.9 mm×15 cm. particle size 3μm column under controlled ambient temperature. Separation among morphine HC1, ketamine HC1, droperidol and their associated degradation compounds was achieved by isocratic elution using mobile phase consisting of methanol/acetonitrile/0.01 M 1-octane sulfonic acid sodium in 0.005 M phosphate buffer (pH 4.0) (50/50/100) at a flow rate of 1.0 ml/mm. An UV/VIS variable programmable wavelength detector set at 254 nm was used. Sample volumes of 20 μL were injected. There was no need for sample pre-treatment.
